Description:
The LACDR, Leiden University Cell Observatory Facility seeks to purchase a highly innovative high content imaging platform where a high-speed confocal microscope integrated with robotic and hotel for imaging multiple well plates in a fully controlled environment. Our research line is focused mainly on the use of human induced pluripotent stem (iPS) cell models for drug research and basic research, and it is crucial to be able to perform the live-cell imaging in a well-controlled incubator as the cell type is highly sensitive for environmental changes. Furthermore, our imaging needs are increasing continuously also due to the higher number of Master students. There is an urgent need for automation to increase our throughput in order to efficiently use the different imaging instruments located in the Cell Observatory. The LIPSI system has all the technical capabilities to meet our scientific requirement. This system will allow to enormously increase our confocal liver cell imaging throughput (hotel with 20 plates) where cells are maintained on the microscope environment in a very stable and controlled incubator, allowing application for sensitive cell types. The platform will be used for both educational and research purpose and for the principal investigators of the Division of Drug Discovery and Safety (DDS): Profs. Bob van de Water, Erik Danen, Micha Drukker, Laura Heitman as well as the head of the HCS facility Dr. Sylvia Le Dévédec. Furthermore, the instrument will be accessible for other PIs of the LACDR who have increased needs to perform advanced imaging. The imaging system will serve Master's students as well in their rotation projects with dedicated professional supervision of the microscopist technician.

For these purposes the high-content imaging platform with integrated automation has to fulfil the following criteria:
1. to image live multiple type of imaging plates automatically in a stable controlled environment
2. can accommodate independent experiments of multiple users simultaneously
3. can combine multiple optical configuration of advanced imaging from different users
4. has high X,Y, Z reloading precision
5. can image fast with high resolution in all kind of plates
6. is sensitive for low fluorescent signal and has low phototoxicity
7. generate data with a format compatible and readable with the data management system Omero

Leiden University has concluded that only the Nikon Lipsi Ti2 A1R Confocal microscope is suitable (see document). This publication is done to inform the market of our intention to award the contract to Nikon Netherlands bv. If no representations are received within 20 days of this publication, the contract will be awarded.
